Mir-612 Inhibits Proliferation and Invasion of Urothelial Carcinoma of Bladder Cells through Activating Hippo Pathway via Targeting PMEPA1

Urothelial carcinoma of bladder (UCB) is a common urological malignancy in the world, but its progression mechanism remains unclear. MiR-612 was found as an anti-tumor factor multiple types cancer, while few studies have revealed functions UCB cells. Based on this, cells such HTB-9 and HTB-4, normal urothelial SV-Huc1, were used subjects this study. Western blot, qRTPCR, CCK-8 assay transwell to assess miR-612 The database, miRWalk, search for potential targets miR-612, dual-luciferase reporter verify accuracy forecasting results. Besides, PMEPA1 overexpressed vector mimics co-transfected into HTB-4 observe regulation miR-612. It that significantly downregulated rather than SV-Huc1 cells, reduced proliferation invasion expression level YAP1 negatively related with LATS1 positively results miRWalk indicated target overexpression could reverse effects including weakened abilities, low high LATS1. Therefore, study suggests inhibits through activating Hippo pathway via targeting PMEPA1.
